The closure takes place on the M1 southbound from J15 to Newport Pagnell Services.
Traffic Officers, Buckinghamshire Fire and Rescue Service and Northamptonshire Fire and Rescue are currently at the scene.
Northamptonshire Fire and Rescue say at 13:29 a crew from Mereway was called to a Mercedes on fire in lane one of the motorway.
Buckinghamshire Fire and Rescue say: "The firefighters used one set of breathing apparatus and one hose reel jet.
"The incident was left in the hands of Thames Valley Police."
UPDATE 2:30PM- National Highways say that recovery are on their way to the area to assist.
UPDATE 3:29PM- Still 2 lanes shut with 1 hour delays southbound J15 to J14 towards MK.
UPDATE 6:00PM- 1 lane to remain closed with the M1 closing overnight for resurfacing.
